The p53 tumor suppressor gene is often mutated in human cancers, leading to loss of its activity. In this study, a p53 gene expression vector was transferred into the OVCAR-3 cells by transfection, and the transfected OVCAR-3 cells (pCDH-p53 OVCAR-3) were studied for sensitivity to chemically induced mouse mammary carcinogenesis. On the basis of numbers of animals needed for reproducible tumor induction, the growth inhibition by pCDH-p53 OVCAR-3 cells was higher than that by OVCAR-3 cells. Histologically, there was no difference between tumors induced by pCDH-p53 OVCAR-3 cells and those induced by OVCAR-3 cells. At the cellular level, p53 protein was expressed in pCDH-p53 OVCAR-3 cells but not in OVCAR-3 cells. These results suggested that p53 gene expression by transfection might induce the growth inhibition of cancer cells.Q:
